What It Is

Epithalon (also spelled Epitalon or Epithalone) is a synthetic tetrapeptide with the sequence alanine-glutamic acid-aspartic acid-glycine, abbreviated Ala-Glu-Asp-Gly or AEDG.[1] It grew out of research on Epithalamin, a bovine pineal-gland extract, largely through the work of Russian researcher Vladimir Khavinson and colleagues on what they termed "peptide bioregulators" -- a research framework, not a recognized pharmaceutical drug class.[2]

Epithalon is a research compound, not a consumer therapeutic. "Epitalon" is the spelling used by PubChem and most PubMed-indexed literature; both names refer to the same molecule.

How It Works

The central hypothesis behind Epithalon is that it can influence telomerase, the enzyme complex that adds repetitive DNA sequences to chromosome ends (telomeres) and helps preserve chromosomal stability during cell division.[3] A widely cited 2003 study reported that Epithalon induced telomerase activity and telomere elongation in human somatic cells.[4]

Separately, related research reported changes in nighttime melatonin rhythm in elderly people and aged monkeys, which is why Epithalon comes up in pineal-signaling and circadian-rhythm discussions.[7] These are proposed mechanisms from laboratory and animal findings, not confirmed human pathways, and telomerase biology is complicated by the fact that the same enzyme is active in many cancers.

What the Research Shows

The strongest evidence for Epithalon is preclinical. Beyond the 2003 telomerase findings, a 2025 study reported telomere-length changes in several human cell lines after Epitalon exposure.[5] Animal studies examining aging biomarkers, lifespan, and tumor incidence in mice found no change in food intake, body weight, or average lifespan, though some aging biomarkers and later-life survival measures were affected.[10]

Human evidence is limited and dated: a small 2002 report described a positive effect in most treated cases of a degenerative retinal condition, and separate studies described melatonin-rhythm changes in elderly subjects and aged monkeys.[9] These studies are small, old, and unreplicated in large modern trials, so they should be read as preliminary leads, not proof that Epithalon treats disease, reverses aging, or extends human lifespan.

Safety & Side Effects

Modern, large-scale human safety data for Epithalon do not exist. FDA's 2026 evaluation did not identify acute or repeat-dose toxicity studies, developmental or reproductive studies, adequate carcinogenicity studies, or clinical studies assessing immunogenicity or aggregation for epitalon free base or acetate.[20]

  • FDA states that it lacks sufficient safety information to know whether compounded Epitalon would cause harm in humans.[14]
  • Aggregation, peptide-related impurities, and immunogenicity remain formulation-dependent concerns.[20]
  • The reported telomerase mechanism creates an unresolved theoretical cancer concern under repeated exposure; FDA found the animal studies inadequate to resolve genotoxic or carcinogenic potential.[20]
  • No established safety profile exists for pregnancy, breastfeeding, long-term use, or medicine interactions.

These gaps mean the absence of reported harm is an absence of adequate surveillance, not a safety guarantee.

Regulatory Status

Epithalon is not FDA-approved for longevity, insomnia, retinitis pigmentosa, or any other use in the United States. An FDA orphan-drug designation record dated September 2, 2010 lists the compound for retinitis pigmentosa but explicitly says "Not FDA Approved for Orphan Indication"; designation is not marketing approval.[13]

At the July 2026 Pharmacy Compounding Advisory Committee meeting, FDA evaluated epitalon free base and acetate for compounded use in insomnia.[15] FDA's briefing proposed not adding either substance to the 503A Bulks List because they were not well characterized and lacked adequate human safety and insomnia-efficacy evidence.[20] That proposal is part of an advisory process rather than an approval, and no compounded or online Epithalon product has FDA-reviewed labeling.

Epithalon's naming is easy to conflate with related terms. Epithalamin is the original crude pineal-gland extract; Epithalon (or Epitalon) is the defined synthetic tetrapeptide studied out of that research line; and Vilon is a separate, related bioregulator peptide from the same research group.[2] Human mortality and longevity claims tied to Epithalamin extract should not be assumed to transfer to synthetic Epithalon unless a study specifically tested it.[19]

Melatonin, the well-characterized pineal hormone, is sometimes mentioned alongside Epithalon since both appear in circadian-rhythm research, but they are distinct molecules with different evidence bases and are not interchangeable.
