Cortagen

Cortagen is a synthetic tetrapeptide with the sequence Ala-Glu-Asp-Pro, abbreviated AEDP, listed in PubChem as H-Ala-Glu-Asp-Pro-OH with molecular formula C17H26N4O9. [1] It belongs to a category of short peptides sometimes called bioregulators, a term used mainly in Russian and Eastern European literature for peptides studied for effects on gene expression and tissue-specific biology. A 2004 paper describing its development states Cortagen was created through directed synthesis based on amino acid analysis of Cortexin, a natural, multi-component peptide preparation from brain cortex tissue. How It Works

Cortagen's proposed mechanism centers on the hypothesis that short peptides can influence gene transcription and protein synthesis, potentially through interactions with chromatin or DNA-associated processes.[3] This comes from the broader peptide-bioregulator research tradition rather than a validated pharmacological model specific to Cortagen.

The main direct evidence is a 2004 microarray study in which mice received daily subcutaneous Cortagen for five days; researchers reported altered expression in 234 of roughly 15,000 examined transcripts in heart tissue.[2] That shows measurable activity in an animal model, but it involved mouse heart tissue, not human brain, and a gene-expression change is not the same as a demonstrated cognitive or neuroprotective benefit.

What the Research Shows

Cortagen's evidence base is overwhelmingly preclinical. In a rat sciatic-nerve-injury model, intramuscular Cortagen increased nerve-fiber growth rate and conduction velocity during regeneration.[4] Mouse studies of locomotor activity and anxiety-related behavior found effects that varied by dose and, at some regimens, appeared anxiogenic rather than calming.[7]

  • Rat and mouse brain-ischemia studies examined related neurospecific peptide preparations for antioxidant and neuroprotective activity[5][6]
  • An in vitro study found Cortagen induced interleukin-2 gene expression less strongly than two related bioregulator peptides[13]
  • A 2015 study of lymphocytes from adults over 80 examined effects on age-related chromatin structure, offering early human-cell context, not a clinical trial[14]

No large, modern human trial of Cortagen was identified. Claims that it improves memory, treats dementia or brain injury, or reverses cognitive decline are unproven extrapolations from animal and cell-based findings.

Safety & Side Effects

Cortagen has no FDA-reviewed label, so there is no standardized adverse-reaction table or contraindication list. Animal research offers limited safety insight: in the mouse behavior studies, effects varied by dose and regimen, and some conditions were associated with increased anxiety-like behavior rather than a clearly benign profile.[7]

Because Cortagen relates to nervous-system and immune-cell signaling, interactions with antidepressants, antiepileptics, sedatives, or immune-modulating medications cannot be ruled out, though none have been formally studied.[3] Compounded or unapproved peptide products also carry quality risks, since they are not reviewed by FDA for purity, sterility, or accurate dosing before reaching the market.[10]

Regulatory Status

No FDA-approved prescribing label for Cortagen was identified in Drugs@FDA or the Orange Book, meaning it has no approved indication, dose, or safety labeling in the United States.[8][9] It should be treated as an investigational research compound, not a regulated medicine, regardless of how it is marketed online.

FDA guidance on compounded drugs notes such products are not evaluated for safety, effectiveness, or quality before marketing, and that poor compounding can cause contamination or incorrect amounts of active ingredient.[10]

Cortagen vs. Cortexin

Cortagen is frequently discussed alongside Cortexin, and the two are related but not interchangeable. Cortexin is a natural, multi-component peptide preparation extracted from brain cortex tissue, while Cortagen is a single, defined synthetic tetrapeptide modeled on one fraction of that extract.[2][7]

Because a complex tissue extract can contain many active components with different receptor targets, findings from Cortexin research, including receptor-binding data reported in rat ischemia studies, should not be assumed to apply to Cortagen.[11] Evidence should be evaluated separately for each compound.
